1. DNA transformation (when necessary)

ii. Cell disruption : binding buffer by the cell lysis,
sonication and centrifugation
iii. Electrophoresis : Samples before supernatant and
centrifugation
iv. When soluble, conduct binding test; for his-tagged
protein of the insoluble,
conduct binding tests using denatured form (using
urea).
